Inadequate aspiration of wells: After pouring washing buffer from the plate, it was
placed/blotted to paper towel to get rid of all the remaining liquid. If these liquid were not
completely eliminated, or if the wells overdried, waiting without doing anything after removing
washing buffer, that could cause problems.
Errors due to experimenter or calibration of pipettes: During experiment, we faced a problem
that caused by the calibration of pipettes. These could cause problems.
The other reason could be reading beyond suggested reading window. Not reading the
absorbance values within time recommended in kit could cause problems but this step was done
by our assistants. Probably there could not be such a kind of error.
Also, in these kind of experiments, regression analysis of the results are recommended by
companies such as R&D Systems. Not using such a kind of data reduction method may cause
less precise fit of the standard curve. Doing such a kind of assay could be better but I do not
think not performing such an analysis could not explain this kind of difference. But if we did
this assay, maybe there will be some small differences between our data.
Not followind incubation times and temperatures: During the experiment, we tried to follows
things written in the kits but there can be some little
differences in
And the last possibility can be inadequate sealing of plate cover which can cause leading to
evaporation.
8-OHdG Microtiter Plate-Precoated With 8-OHdG was used to see the effect of
concentrationand absorbance values. To do this samples containing 8-OHdG was used. Primary
antibody monoclonal antibody is specific to 8-OHdG because plate is precoated with 8-OHdG.
Secondary antibody which is HPR-conjugated were used and with the aid of this enzyme,
chromogen was cleaved and this reaction gave yellow color.

Dünya standardında ölçüler, metrik sistem kulanılarak yapılır. Dünya çapında bilim
insanlarının kullanmasının yanında, neredeyse bütün ülkeler metrik sistemi standart ölçü
birimleri olarak kullanırlar. Bu dersimizdeki tüm ölçümler metrik sistem kullanılarak
yapılacaktır.
